During clathrin-mediated endocytosis, a small patch of flat plasma membrane is invaginated into the cell and form a vesicle under the action of a clathrin coat. How the clathrin coat deforms the membrane remains unclear and two main hypotheses have been debated. The “constant area” hypothesis assumes that clathrin molecules initially form a flat lattice which gradually obtains intrinsic curvature and finally is bent into a spherical cap while keeping the area of the clathrin lattice constant. The alternative “constant curvature” hypothesis assumes that the intrinsic curvature of the clathrin lattice remains constant while the surface area it covers increases. In this talk, I will present our recent work on the vesiculation pathway of clathrin-mediated endocytosis. By applying the Helfrich model of membrane, we were able to calculate the membrane shape for different coating area and spontaneous curvature of the clathrin coat, as well as the corresponding free energy. We found that vesiculation could happen as long as the coating area or the spontaneous curvature is large enough. We then constructed the assembling pathway of the clathrin-coat by applying the gradient-descend method to the free energy landscape of the membrane. We found that the assembling pathway is neither the constant area model nor the constant curvature model but a combination of both. By comparing with experimental data, we found that our model fit better than the two existing models.
